How do they even track the variants with the same test when it has never been isolated?

So, a physician in Africa says she had patients with mild common cold symptoms that alarmed her that maybe it was a new Covid variant, and led to discovery of new variant . . . say what?! They have never isolated Covid, but they can isolate new variants . . . say what?! Withing 3-4 days of disovery, they miraculously have a test to identify the new variant . . . say what?! Not only do they have the new test for the new variant, they have it available all over the world . . . say what?! Then, a plane leaves South Africa where all passengers were tested for Covid before boarding and were negative. The plane lands in Johanesburg and 10% of the passengers miraculously have the Omicron variant (tests beat them there from South Africa). . . say what?! Of course, by the time the plane landed, the world had also determined the Omicron variant is more infectious than other variants; children particuallry susceptible; and the vaccines does not provide protection against Omicron. Wow!!! Say what?! Of course, the pharma companies also altready determined they could come out with a new vaccine targeting Omicron in 30-60 days. . . say what?! I could go on, but the fuckery is simply never ending.
